Recent studies on histamine receptor (HR) subtypes identified imidazolyl butyl cyanoguanidines, like UR-PI376, as highly potent agonists at the human histamine H-4 receptor (hH(4)R). Investigations in radioligand-binding assays exhibited only very weak activity at the hH(1)R and hH(3)R, while nearly all compounds were inactive at the hH(2)R and hH(4)R. Seleno-Michael Reaction of Stable Functionalised Alkyl Selenols: A Versatile Tool for the Synthesis of Acyclic and Cyclic Unsymmetrical Alkyl and Vinyl Selenides

Seleno-Michael additions of stable functionalised primary alkyl selenols to activated alkenes and alkynes are described. In the presence of Al2O3, -hydroxy-, -amino-, and -mercapto selenols react smoothly with electron-poor alkenes and alkynes to afford the corresponding unsymmetrical functionalised dialkyl- and alkyl-vinyl-selenides in good yield. The very mild conditions allow a broad range of selenols and Michael acceptors to be converted into the corresponding synthetically valuable seleno-Michael adducts, demonstrating high selectivity and excellent functional group tolerance. Hydroxy- and mercapto-substituted vinyl selenides were efficiently employed for the synthesis of functionalised 1,3-oxaselenolanes, 1,3-thiaselenolanes, and 1,4-thiaselenanes through intramolecular oxa- and thia-Michael additions. Furthermore, a NaH-promoted lactonization enables the synthesis of variously substituted 2-oxo-1,4-oxaselenanes from hydroxy-vinyl-selenides. Evaluation of thiol peroxidase-like properties of novel functionalised organoselenides demonstrated that they possess a remarkable catalytic antioxidant activity.

New optimized piperamide analogues with potent in vivo hypotensive properties

We describe herein the structural optimization of new piperamide analogues, designed from two natural prototypes, piperine 1 and piper-dardine 2, obtained from Piper tuberculatum Jacq. (Piperaceae). Molecular modeling studies using semiempirical AMI method were made in order to establish rational modifications to optimize them by molecular simplification. The targeted compounds (10) and (11) were respectively obtained using benzaldehyde (12) and para-anisaldehyde (13) as starting materials. Design, Synthesis and Study of Nitrogen Monoxide Donors as Potent Hypolipidaemic and Anti-Inflammatory Agents

Inflammation and oxidative stress are involved in cardiovascular diseases. Nitrogen monoxide participates in the regulation of endothelial processes. Thus, derivatives of classic nonsteroidal anti-inflammatory drugs (NSAIDs), trolox or cinnamic acids esterified with 2-(nitrooxy)ethanol were designed and studied. It was found that the nitrogen monoxide (NO) releasing activity was comparable to that of S-nitroso-N-acetylpenicillamine. The nitrooxy derivatives decreased potently lipid indices in the plasma of hyperlipidaemic rats (30-85%). All compounds presented increased anti-inflammatory activity in vivo, inhibiting carrageenan-induced rat paw oedema as high as 76%, up to six times higher than that of the parent acids. Lipoxygenase inhibitory activity was significant for most of them, although the parent molecules exerted a minor effect (IC50 > 0.2 mM). Those compounds incorporating an antioxidant structure inhibited rat microsomal membrane lipid peroxidation strongly and possessed radical scavenging activity. These results indicated that the described compounds could act at different targets in multifactorial diseases, further limiting the possible adverse effects of drug combinations.

Second-order rate constants k(1)(298) are 1.61 x 10(4), 8.0 x 10(4), 3.79 x 10(4), 3.69 x 10(4), 2.21 x 10(4), 1.84 x 10(4), 1.91 x 10(3), 1.34 x 10(4), 1.52 x 10(4), 7.75 x 10(3), 900, and 5.2 x 10(3) M(-1) s(-1), respectively. No steric threshold is observed for these sterically unhindered systems. There is a rough compensation effect between Delta H-1 double dagger and Delta S-1 double dagger, i.e. a smaller Delta H-1 double dagger is usually accompanied by a larger negative Delta S-1 double dagger, indicating that all thioethers react via the same mechanism. It appears that the much lower reactivity observed for the highly branched (t-Bu)(2)S is primarily caused by a high activation enthalpy. Volumes of activation have been determined for a series of thioethers with a constant cone angle, viz. EtSCH(2)COOH, S(CH2COOH)(2), and S(CH2CH2COOH)(2) through high-pressure stopped-flow measurements. Values of Delta V-1 double dagger are -7.9 +/- 0.5, -8.1 +/- 0.4, and -7.6 +/- 0.3 cm(3) mol(-1), respectively. These values together with that for Et(2)S (-8.7 +/- 0.1 cm(3) mol(-1)) determined previously shows that variation of electronic properties, steric factors being kept constant, change the reactivity markedly, but have no observable influence on the activation volumes. Stability constants beta(1) for five palladium thioether complexes derived as the ratio between rate constants for forward and reverse reactions vary between (1.2 +/- 0.3) x 10(4) and (3.2 +/- 0. 7) x 10(4) M(-1).
