Identification of new derivatives of sinigrin and glucotropaeolin produced by the human digestive microflora using H-1 NMR spectroscopy analysis of in vitro incubations

One- and two-dimensional H-1 NMR spectroscopy were used to study the biotransformation of two dietary glucosinolates, sinigrin (SIN), and glucotropaeolin (GTL) by the human digestive microflora in vitro. The molecular structures of the new metabolites issued from the aglycone moiety of the glucosinolate were identified, and the modulation of carbon metabolism was studied by quantifying bacterial metabolites issued from the xenobiotic incubation in the presence or absence of a source of free glucose. Unambiguously and for the first time, it was shown that SIN and GTL were transformed quantitatively into allylamine and benzylamine, respectively. The comparison of the kinetics of transformation of SIN and GTL with and without glucose clearly showed that the presence of glucose did not modify either the nature of the metabolites or the rate of transformation of the glucosinolates (complete degradation within 30 h). The main end products of the glucose moiety of glucosinolates were characteristic of anaerobic carbon metabolism in the digestive tract (acetate, lactate, ethanol, propionate, formate, and butyrate) and similar to those released from free glucose. This work represents the first application of H-1 NMR spectroscopy to the study of xenobiotic metabolism by the human digestive microflora, demonstrating allyl- and benzylamine production from glucosinolates. Whether these amines are produced in vivo from dietary glucosinolates remains to be established. This would reduce the availability of other glucosinolate metabolites, notably cancer-protective isothiocyanates.

New Cyclohexylamine-dithiocarbamate Derivatives as Potential Anti-microbial Agents

Background: In this study, 2-(substituted-sulfanyl)-N, N-dicyclohexylacetamide derivatives (2a-2g) and 2-(dicyclohexylamino)-2-oxoethyl-1-substituted carbodithioate derivatives (2h-2m) were synthesized and screened for their antimicrobial activity. Methods: Newly synthesized compounds were screened against two gram negative bacteria (S. typhimurium and E. coli), three gram positive bacteria (S. aureus, B. cereus and L. monocytogenes), four Candida species, four Aspergillus spp. and three Penicillium spp. Among them (2a-2m), compounds 2i (2-(dicyclohexylamino)-2-oxoethyl-thiomorpholine-4-carbodithioate) and 2k (2( dicyclohexylamino)-2-oxoethyl-4-(4-methoxyphenyl) piperazine-1-carbodithioate) were detected to have higher inhibitory effect than other compounds. Results and Conclusion: Minumum inhibitor concentrations (MICs) of the compounds were determined between the range of 97.5-390 mu g/mL. Additionally, parameters determined that some physicochemical and toxic properties were predicted using computational methods.

DESIGN, SYNTHESIS AND ANTIMICROBIAL ACTIVITIES OF SOME AZOLE DERIVATIVES

Three new 1,3,4-oxadiazole, 1,3-thiazolidine and 1,2,4-triazole derivatives were obtained starting from furan-2-carbohydrazide. Then, 1,2,4-triazole compound was converted to the corresponding Mannich bases using several secondary amines including piperidine, piperazine, morpholine or thiomorpholine moiety. The synthesis of 5-(furan-2-yl)-4-{[(4-methoxyphenyl)methylidene]amino}-4H-1,2,4-triazole-3-thiol (XIII) was performed starting from furan-2-carbohydrazide by three steps. The structures of the synthesized compounds were well characterized by elemental analyses, IR, H-1 NMR, C-13 NMR and mass spectral studies. Newly synthesized compounds were screened for their antimicrobial activities and some of them displayed activity against the tested microorganisms.

Functionalization of Ruthenium(II)(eta(6)-p-cymene)(3-hydroxy-2-pyridone) Complexes with (Thio)Morpholine: Synthesis and Bioanalytical Studies

Herein, [Ru-II(eta(6)-p-cymene)] complexes with 3-hydroxy-2-pyridinone functionalized with morpholine and thiomorpholine, as a means often used in medicinal chemistry to alter the physicochemical properties of drug compounds, are reported. The compounds underwent hydrolysis of the Ru-Cl bond and the aqua species were stable for up to 48 h in aqueous solution, as observed by H-1 NMR spectroscopy and ESI-MS. The compounds formed adducts with amino acids and proteins through cleavage of the pyridinone ligand. Binding experiments to the nucleosome core particle by means of X-ray crystallography revealed similar reactivity and exclusive binding to histidine moieties of the histone proteins. Preliminary cyclin-dependent kinase 2 (CDK2)/cyclin A kinase inhibitory studies revealed promising activity similar to that of structurally related organometallic compounds.

Carbamate formation is one of the major chemical reactions that can occur in solution in the capture of CO2 by amine-based solvents, and carbamate formation makes a significant enthalpy contribution to the absorption-desorption of CO2 that occurs in the absorber/stripper columns of the PCC process. Consequently, the formation of carbamates of selected series of primary and secondary amines over the temperature range (288 to 318) K has been investigated by equilibrium H-1 NMR studies, and the stability constants (K-9) for the equilibrium: RNH2 + HCO3- reversible arrow(K9) RNHCOO- + H2O are reported. van’t Hoff analyses have resulted in standard molar enthalpies, Delta H-m(o), and entropies, Delta S-m(o), of carbamate formation. A Delta H-m(o) – Delta S-m(o) plot generates a linear correlation for carbamate formation (providing a mean standard molar free energy, Delta G(m)(o), for carbamate formation of about -7 kJ . mol (1)), and this relationship helps provide a guide to the selection of an amine(s) solvent for CO2 capture, in terms of enthalpy considerations. A linear Delta H-m(o) – Delta S-m(o) plot also occurs for carbamate protonation. The formation of the carbamates has been correlated with systematic changes in composition and structure, and steric effects have been identified by comparing molecular geometries obtained using density functional B3LYP/6-311++G(d,p) calculations. Trends in steric effects have been identified in the series of compounds monoethanolamine (MEA), 1-amino-2-propanol, 2-amino-1-propanol (AP) and 2-amino-2-methyl-1-propanol (AMP). In the case of 2-piperidinemethanol, 2-piperidineethanol and 3-piperidinemethanol, strong intramolecular hydrogen bonding is shown to be the likely cause for lack of carbamate formation, and in the ring systems of pyrrolidine, morpholine, piperidine and thiomorpholine trends in carbamate formation (as given by K-9) have been correlated with the internal ring angle at the amine nitrogen, as well as the planarity of the environment around the nitrogen atom. Degradation of morpholine and thiomorpholine by an environmental Mycobacterium involves a cytochrome P450. Direct evidence of intermediates by in situ H-1 NMR

A strain of Mycobacterium sp. RP1 was isolated from a contaminated activated sludge. It was capable of utilizing morpholine, a waste of chemical industry, as sole source of carbon, nitrogen and energy. The kinetic of biodegradation of morpholine was followed directly on the incubation medium using in situ H-1 NMR. This technic allowed to identify two intermediates of the degradative pathway: glycolate and 2-(2-aminoethoxy)acetate. The inhibitory effects of metyrapone on the degradative abilities of the strain RP1 indicated the involvement of a cytochrome P450. This observation was confirmed by spectrophotometric analysis and H-1 NMR. Metyrapol, a known metabolite of metyrapone, was also found to be an inhibitor. Syntheses and properties of mixed dinuclear copper(II) complexes with heterocyclic dithiocarbamates and a cyclic octadentate tertiary amine

Five new Cu-II complexes of general formula [Cu-2(Rdtc)tpmc](ClO4)(3), (1)-(5), where tpmc and Rdtc(-) refer to N,N’,N,N’-tetrakis(2-pyridylmethyl)-1,4,8,11-teraazacyclotetradecane and piperidine- (Pipdtc), 4-morpholine-(Morphdtc), 4-thiomorpholine- (Timdtc), piperazine- (Pzdtc) or N-methylpiperazine- (N-Mepzdtc) dithiocarbamates, respectively, have been prepared. Elemental analyses, conductometric and magnetic measurements, u.v./vis, i.r., e.p.r. and mass spectroscopy have been employed to characterize them. The complexes adopt an exo coordination of CuII ions and tpmc. The dithiocarbamate ion joins both the sulphur and the copper atoms acting as a bridging ligand The presence of different heteroatoms in the piperidine ring influences the nu(Cdouble bondN) and nu(Cdouble bondS) vibrations which decrease in the order of the complexes: Pipdtc > N-Mepipdtc > Pzdtc > Morphdtc > Timdtc ligands. Attention has been paid to the detailed mechanism of the mass spectral fragmentation of the complexes. The g(eff) factors of the complexes have been also estimated by e.p.r. spectra. Finally, the complexes obtained demonstrate microbiologycal activity against some bacteria.
