A Metabolomics Study of Retrospective Forensic Data from Whole Blood Samples of Humans Exposed to 3,4-Methylenedioxymethamphetamine: A New Approach for Identifying Drug Metabolites and Changes in Metabolism Related to Drug Consumption

The illicit drug 3,4-methylenedioxymethamphetamine (MDMA) has profound physiological cerebral, cardiac, and hepatic effects that are reflected in the blood. Screening of blood for MDMA and other narcotics are routinely performed in forensics analysis using ultra performance liquid chromatography with high-resolution time-of-flight mass spectrometry (UPLC-HR-TOFMS). The aim of this study was to investigate whether such UPLC-HR-TOFMS data collected over a two-year period could be used for untargeted metabolomics to determine MDMA metabolites as well as endogenous changes related to drug response and toxicology. Whole blood samples from living Danish drivers’ positive for MDMA in different concentrations were compared to negative control samples using various statistical methods. The untargeted identification of known MDMA metabolites was used to validate the methods. The results further revealed changes of several acylcarnitines, adenosine monophosphate, adenosine, inosine, thiomorpholine 3-carboxylate, tryptophan, S-adenosyl-L-homocysteine (SAH), and lysophospatidylcholine (lysoPC) species in response to MDMA. These endogenous metabolites could be implicated in an increased energy demand and mechanisms related to the serotonergic syndrome as well as drug induced neurotoxicity. In aqueous solution, macrocyclic ligand cyclam is characterized by the anodic peak at 0.95 V. The Rdtc(-) ligands have two anodic peaks, one in the region 0.25-0.30 V and the other in the 0.78-0.95 V region. Absence of these anodic peaks in the case of the complexes indicates that coordination to cobalt(III) stabilizes both cyclam and Rdtc(-) ligands, but reversible peaks in the range -0.68 to -0.78 V support the Co(III) redox reaction. Identification of a novel oxazolidinone (U-100480) with potent antimycobacterial activity

During the course of our investigations in the oxazolidinone antibacterial agent area, we have identified a subclass with especially potent in vitro activity against mycobacteria. The salient structural feature of these oxazolidinone analogues, 6 (U-100480), 7 (U-101603), and 8 (U-101244), is their appended thiomorpholine moiety. The rational design, synthesis, and evaluation of the in vitro antimycobacterial activity of these analogues is described. Potent activity against a screening strain of Mycobacterium tuberculosis was demonstrated by 6 and 7 (minimum inhibitory concentrations or MIC’s less than or equal to 0.125 mu g/mL). A Delta H-m(o) – Delta S-m(o) plot generates a linear correlation for carbamate formation (providing a mean standard molar free energy, Delta G(m)(o), for carbamate formation of about -7 kJ . mol (1)), and this relationship helps provide a guide to the selection of an amine(s) solvent for CO2 capture, in terms of enthalpy considerations. Herein, we report the reactions of 1,1,2,4,4-pentachloro-3-nitro-1,3-butadiene 1a and (1Z)-1-bromo-1,2,4,4-tetrachloro-3-nitro-1,3-butadiene 1b with nitrogen- and sulfur-containing nucleophiles to obtain highly functionalized S-, S,S-, S,S,S-, S,O- and N,S-substituted-polyhalodiene-3-nitro-1,3-butadiene derivatives.
