In the context of increasing drug resistance among Mycobacterium tuberculosis (Mtb) strains, new, potent and well tolerated drugs are urgently needed. Linezolid, the first and only marketed oxazolidinone antibiotic, has shown certain activity against Mtb. However, serious safety concerns related to long-term administration have limited its utility for TB treatment. Sutezolid (formerly PNU-100480) is a thiomorpholine analogue of linezolid with greater potency in several experimental TB models and the potential for a superior safety profile. This article reviews available preclinical and clinical data regarding the pharmacokinetics, safety and efficacy of sutezolid and presents the case for its further development for the treatment of TB.
