One-pot synthesis of isoindole fused quinazolin 4-ones via intramolecular 1,3 hydride transfer in the presence of acid catalyst has been described. Substrate scope and mechanistic insights were investigated. Substituents on the amide side have a negligible influence on the key step and therefore the method have wide scope for accessing various bicyclic core structure.

Synthesis of pyrazole-3-carboxylic acid was progressed via two different protocols, one of which is solid state. Pyrazole-3-carboxylic acid was converted into different derivatives such as ester, urea, amide and nitrile. The amide compound was converted to nitrile using SOCl2 and DMF. Solid state heating of carboxylic acid gave decarboxylated product. Cyclization of tetra-substituted pyrazole with hydrazines resulted in pyrazolopyridazinones. The antimicrobial activities of the synthesized pyrazole derivatives against Bacillus cereus, Escherichia coli, Micrococcus luteus, Staphylococcus aureus, and Saccharomyces cerevisiae were evaluated. One of the pyrazole derivatives which possess nitro group showed antimicrobial activity in only B. cereus, a Gram-positive bacteria, with an MIC of 128 mu g/mL.

fac-[Re(CO)(3)(kappa(2)-N-im,S-bptsc)Cl] (2), isolated from the reaction between Re(CO) 5 Cl and benzophenone thiosemicarbazone, bptsc, (1) in refluxing toluene in air, is the first Re compound of 1 and is the second Re(CO)(3)Cl compound of kappa(2)-N-im,S-coordinated ligand (im = imine). The authenticity of 2 was established from the results of its elemental composition, spectroscopic measurements, and X-ray crystallographic analysis. Single crystals of 2 grown from DMF are in the monoclinic space group P2(1)/c. The asymmetric unit of 2 revealed pseudo-octahedral coordination about Re. Two carbonyl C atoms, an imine N atom and a thione S atom occupy the equatorial sites and the axial sites are occupied by a carbonyl C atom and a Cl atom. The kappa(2)-N,S-coordination of 1 to Re forms a semi-planar five-membered [Re-N-N-C-S] metallocyclic ring. The extended structure disclosed stacks of molecules interlocked via a web of N-H center dot center dot center dot X (X = Cl or O), S center dot center dot center dot S and C-H center dot center dot center dot pi interactions. DFT calculations divulged facile delocalization of electron density in the molecule and interatomic distances and angles in good agreement with the solid state structure. Electrochemical measurements on 2 in CH3CN and DMF revealed sequential irreversible electron transfers pointing to structural changes due to electrochemically induced thione-thiol tautomerization of the thioamide moiety. Plausible mechanisms for the oxidative and reductive electrochemical decomposition of 1 and 2 are reported. Low Magnetic field effects on a photoinduced electron transfer reaction between benzophenone and 1,4-diazabicyclo [2.2.2] octane (DABCO) in an ionic liquid of N,N,N-trimethyl-N-propylammonium bis(trifluoromethanesulfonyl)amide were studied with nanosecond transient absorption measurements. At a magnetic field (similar to 2 mT) comparable with the hyperfine coupling in DABCO cation, so-called low field effects (LFEs), whose effect is opposite to the magnetic field effects caused by the hyperfine coupling mechanism, were observed on the yield of the benzophenone anion radical. The magnitude of the LFE is linearly increased with the increase of the lifetime of the radical ion pair. The present study provides basic data to clarify the mechanism of LFEs in biological systems.

Purification of recombinant proteins is often a challenging matter because high purity and high recovery are desired. If the expressed recombinant protein is also in a complex matrix, such as from the silkworm expression system, purification becomes more challenging. Even if purification from the silkworm expression system is troublesome, it benefits from a high capacity for the production of recombinant proteins. In this study, magnetic nanoparticles (MNPs) were investigated as a suitable tool for the purification of proteins from the complex matrix of the silkworm fat body. The MNPs were modified with nickel so that they have an affinity for His-tagged proteins, as the MNP purification protocol itself does not need special equipment except for a magnet. Among the three different kinds of investigated MNPs, MNPs with sizes of 100 nm to 200 nm and approximately 20 nm-thick nickel shells were the most suitable for our purpose. With them, the total protein amount was reduced by up to at least approximately 77.7%, with a protein recovery of around 50.8% from the silkworm fat body. The minimum binding capacity was estimated to be 83.3 mu g protein/mg MNP. Therefore, these MNPs are a promising tool as a purification pretreatment of complex sample matrices.

The development of metal phosphide catalysts for organic synthesis is still in its early stages. Herein, we report the successful synthesis of single-crystal cobalt phosphide nanorods (Co2P NRs) containing coordinatively unsaturated Co-Co active sites, which serve as a new class of air-stable, highly active, and reusable heterogeneous catalysts for the reductive amination of carbonyl compounds. The Co2P NR catalyst showed high activity for the transformation of a broad range of carbonyl compounds to their corresponding primary amines using an aqueous ammonia solution or ammonium acetate as a green amination reagent at 1 bar of H-2 pressure; these conditions are far milder than previously reported. The air stability and high activity of the Co2P NRs is noteworthy, as conventional Co catalysts are air-sensitive (pyrophorous) and show no activity for this transformation under mild conditions. P-alloying is therefore of considerable importance for nanoengineering air-stable and highly active non-noble-metal catalysts for organic synthesis.

Group 6 alkynyl Fischer carbene complexes behave as electrophiles with highly reactive masked dienes such as anthrone and 3-hydroxy-2-pyrone, which also can react as bidentate nucleophiles. Both Cr-0 and W-0 alkynyl carbene complexes require tertiary amines to promote the formation of 1,4-C-addition products, but a marked dependence of the reactivity on the metal was observed. Although W-0 complexes form exclusively the 1,4-C-adducts in the presence of a catalytic amount of base, Cr-0 complexes lead to new anthrone-derived bis-carbene complexes, obtained by a sequential 1,4-C- and 1,4-O-addition process with high diastereoselectivity and only in the presence of an excess of base. We report a set of electrochemically regulated protocols for the divergent synthesis of ketones and beta-keto esters from the same beta-hydroxycarboxylic acid starting materials. Enabled by electrochemical control, the anodic oxidation of carboxylic acids proceeded in either a one-electron or a two-electron pathway, leading to a 1,4-aryl transfer or a semipinacol-type 1,2-group transfer product with excellent chemoselectivity. The 1,4-aryl transfer represents an unprecedented example of carbon-to-oxygen group transfer proceeding via a radical mechanism. In contrast to previously reported radical group transfer reactions, this 1,4-group transfer process features the migration of electron-rich aryl substituents. Furthermore, with these chemoselective electrochemical oxidation protocols, a range of ketones and beta-keto esters including those possessing a challenging-to-access medium-sized ring could be synthesized in excellent yields.

Due to the redox properties closely related to numerous physiological and pathological processes, biothiols, including cysteine (Cys), homocysteine (Hcy) and glutathione (GSH), have received considerable attention in biological science. On account of the important physiological roles of these biothiols, it is of profound significance to develop sensitive and selective detection of biothiols to understand their biological profiles. In this work, we reported an efficient fluorescent probe, PHPQ-SH, for detecting biothiols in vitro and vivo, based on the phenothiazine-HPQ skeleton, with DNBS (2,4-dinitrobenzenesulfonate) as the response unit. Probe PHPQ-SH exhibited brilliant sensing performances toward thiols, including a large Stokes shift (138 nm), excellent sensitivity (for GSH, LOD = 18.3 nM), remarkable fluorescence enhancement (163-fold), low cytotoxicity, rapid response (8 min), and extraordinary selectivity. Finally, the probe PHPQ-SH illustrated herein was capable of responding and visualizing biothiols in MCF-7 cells and zebrafish.

Objective: Coronavirus disease 2019 (COVID-19) is associated with abnormal inflammatory and coagulation markers, potentially mediating thrombotic events. Our objective was to investigate the incidence, time course, laboratory features, and in-hospital outcomes of COVID-19 patients with suspected venous thromboembolism (VTE). Methods: A retrospective observational cohort study was conducted of patients hospitalized with COVID-19 who had undergone ultrasound imaging for suspected VTE from March 13 to May 18, 2020. The medical records of the included patients were reviewed for D-dimer, fibrinogen, prothrombin time, partial thromboplastin time, platelet count, C-reactive protein (CRP), and high-sensitivity troponin T at admission and at up to seven time points before and after ultrasound examination. The clinical outcomes included superficial venous thrombosis, deep vein thrombosis, pulmonary embolism, intubation, and death. Mixed effects logistic, linear, and Cox proportional hazards methods were used to evaluate the relationships between the laboratory markers and VTE and other in-hospital outcomes. Results: Of 138 patients who had undergone imaging studies, 44 (31.9%) had evidence of VTE. On univariable analysis, an elevated admission CRP (odds ratio [OR], 1.05; 95% confidence interval [CI], 1.01-1.09; P = .02; per 10-U increase in CRP), platelet count (OR, 1.48; 95% CI, 1.04-2.12; P = .03; per 1000-U increase in platelet count), and male sex (OR, 2.64; 95% CI, 1.19-5.84; P = .02), were associated with VTE. However only male sex remained significant on multivariable analysis (OR, 2.37; 95% CI, 1.01-5.56; P = .048). The independent predictors of death included older age (hazard ratio [HR], 1.04; 95% CI, 1.00-1.07; P = .04), active malignancy (HR, 4.39; 95% CI, 1.39-13.91; P = .01), elevated admission D-dimer (HR, 1.016; 95% CI, 1.003-1.029; P = .02), and evidence of disseminated intravascular coagulation (HR, 4.81; 95% CI, 1.76-13.10; P = .002). Conclusions: Male sex, elevated CRP, and elevated platelet count at admission were associated with VTE on univariable analysis. However, only male sex remained significant on multivariable analysis. Older age, active malignancy, disseminated intravascular coagulation, and elevated D-dimer at admission were independently associated with death for patients hospitalized with COVID-19.
