The one-pot synthesis of quinazoline-2,4-diones was developed in the presence of 4-dimethylaminopyridine (DMAP) by metal-free catalysis. The commercially available (Boc)(2)O acted as a key precursor in the construction of the 2-position carbonyl of quinazolinediones. The p-methoxybenzyl (PMB)-activated hetero- cyclization could smoothly proceed at room temperature instead of the microwave condition. This strategy is compatible with a variety of substrates with different functional groups. Furthermore, this protocol was utilized to smoothly prepare Zenarestat with a total yield of 70%.

The utilization of photochemical reaction channel based on radical process is rarely reported, which might be a very efficient and feasible strategy for improving generation of Type I reactive oxygen species (ROS). In this work, a double ionic-type aggregation-induced emission luminogen (AIEgen) of TIdBO was developed as a photosensitizer, of which the potential photocyclization characteristic involving an electron-transfer process had a positive effect on Type I ROS generation in aggregates under continuous light irradiation. Its noticeable photodynamic therapy (PDT) performance and self-monitoring of PDT process by the relationship between cellular morphology change and fluorescence intensity enhancement were achieved. In addition, it showed a good killing ability to microbes and specific interactions with microbes but not cells by regulating the incubation time. These intriguing results reveal a feasible design principle for the implementation of efficient PS preparation in clinical treatment under hypoxic conditions.

Herein, we disclose a unique directing effect of 9-substituted triptycenes in electrophilic substitution to achieve the regioselective functionalization of the triptycene core. The Hirshfeld population analysis was adopted to predict the selectivity in electrophilic substitution. TMS and t-Bu groups were found to considerably accelerate the reaction at C2 positions to produce C3symmetric isomers. Correlation between distortion and charge distribution within benzene rings was systematically examined.

We report the synthesis of inexpensive and environmentally benign cobalt(0) nanoparticles on L-3,4-dihydroxyphenylalanine (L-dopa) functionalized and magnetite (Fe3O4) grafted graphene oxide (Co@GO/Fe3O4/L-dopa) which was fully characterized with different techniques such as Scanning Electron Microscopy (SEM), High Resolution Transmission Electron Microscopy (HR-TEM), X-Ray Photoelectron Spectroscopy (XPS), X-ray Powder Diffraction (XRD), Thermogravimetric analysis (TGA), Fourier Transform Infrared Spectroscopy (FTIR), Vibrating Sample Magnetometer (VSM), Carbon Hydrogen Nitrogen (CHN) analysis, Energy Dispersive X-ray (EDX) and Inductively Coupled Plasma Atomic Emission Spectroscopy (ICP-AES). Graphene oxide was used as the core material due to its mechanical, electrical and thermal properties. In order to avoid the cumbersome process of separation, magnetite nanoparticles were coated over the graphene oxide. After the successful preparation of graphene oxide based magnetic nanoparticles, L-dopa was grafted over Fe3O4 nanoparticles so as to provide firm anchoring agent for cobalt nanoparticles. Finally, cobalt(0) nanoparticles were immobilized on the developed magnetic support. The catalytic efficiency of the synthesized catalyst was tested for Suzuki cross-coupling and oxidation reactions, usually carried out by precious and expensive second and third row transition metals; products were obtained in good to excellent yields. The synthesized catalyst represents an attractive alternative to conventional catalysts for Suzuki cross-coupling and oxidation reactions, and is recyclable up to five runs.

The (R)- and (S)-enantiomers of the chiral herbicide napropamide (NAP) show different biological activities and ecotoxicities. These two enantiomers behave differently in the environment due to enantioselective catabolism by microorganisms. However, the molecular mechanisms underlying this enantioselective catabolism remain largely unknown. In this study, the genes (snaH and snpd) involved in the catabolism of NAP were cloned from Sphingobium sp. B2, which was capable of catabolizing both NAP enantiomers. Compared with (R)-NAP, (S)-NAP was much more rapidly transformed by the amidase SnaH, which initially cleaved the amide bonds of (S)/(R)-NAP to form (S)/(R)-2-(1-naphthalenyloxy)-propanoic acid [(S)/(R)-NP] and diethylamine. The alpha-ketoglutarate-dependent dioxygenase Snpd, showing strict stereoselectivity for (S)-NP, further transformed (S)-NP to 1-naphthol and pyruvate. Molecular docking and site-directed mutagenesis analyses revealed that when the (S)-enantiomers of NAP and NP occupied the active sites, the distance between the ligand molecule and the coordination atom was shorter than that when the (R)-enantiomers occupied the active sites, which facilitated formation of the transition state complex. This study enhances our understanding of the preferential catabolism of the (S)-enantiomer of NAP on the molecular level.

The thermochemical network of 91 isodesmic and other balanced reactions was used to adjust simultaneously the enthalpies of formation of 14 urea compounds. The experimental enthalpies of formation of these compounds were treated as unknowns in the least-squares minimization of deviations between the theoretically predicted enthalpies of reaction and experimental values. A result of a least-squares fit confirms the accuracy of experimental enthalpies of formation of 8 ureas, whereas the experimental measurements for 6 urea compounds require re-examination. The gas-phase standard enthalpies of formation of 8 ureas were used as a consistent basis to obtain values for other 15 ureas through isodesmic reactions. Although the synthesis of novel N-heterocyclic molecules is extremely demanding as well as challenging, the involvement of toxic solvents often triggers environmental safety concerns, resulting in process-engineering challenges. Herein, we have demonstrated a rapid, environmentally benign and energy efficient scalable method for the synthesis of 2,3-dihydroquinazolin-4(1H)-one by grinding in a mortar pestle as well as mechanochemically via ball milling using p-TSA catalyst. The ability to accomplish the reaction in the absence of solvent via grinding or milling with p-TSA catalyst, with an immediate reduction in the cost and operational procedures, features the significant advantages of this protocol. The scalability and significance of the operational parameters during mechanochemical milling in the tubular ball mill were also demonstrated. Excellent yield in short duration, large substrate scope, product scalability, and easy recoverability are the prime features of this mechanochemical solvent-free protocol for the synthesis of 2,3-dihydroquinazolin-4(1H)-one. The study also demonstrated the significant role of ball diameter to improve the efficiency of the milling operation in this mechanochemical synthesis.

OBJECTIVE Neurosurgical education in the US has changed significantly as a consequence of the novel coronavirus (COVID-19) pandemic. Institutional social distancing requirements have resulted in many neurosurgical programs utilizing video conferencing for educational activities. However, it is unclear how or if these practices should continue after the pandemic. The objective of this study was to characterize virtual education in neurosurgery and understand how it should be utilized after COVID-19. METHODS A 24-question, 3-part online survey was administered anonymously to all 117 US neurosurgical residency programs from May 15, 2020, to June 15, 2020. Questions pertained to the current use of virtual conferencing, preferences over traditional conferences, and future inclinations. The Likert scale (1 = strongly disagree, 3 = neutral, 5 = strongly agree) was used. Comparisons were calculated using the Mann-Whitney U-test. Statistical significance was set at 0.05. RESULTS One-hundred eight responses were recorded. Overall, 38 respondents (35.2%) were attendings and 70 (64.8%) were trainees. Forty-one respondents (38.0%) indicated attending 5-6 conferences per week and 70 (64.8%) attend national virtual conferences. When considering different conference types, there was no overall preference (scores < 3) for virtual conferences over traditional conferences. In regard to future use, respondents strongly agreed that they would continue the practice at some capacity after the pandemic (median score 5). Overall, respondents agreed that virtual conferences would partially replace traditional conferences (median score 4), whereas they strongly disagreed with the complete replacement of traditional conferences (median score 1). The most common choices for the partial replacement of tradition conferences were case conferences (59/108, 55%) and board preparation (64/108, 59%). Lastly, there was a significant difference in scores for continued use of virtual conferencing in those who attend nationally sponsored conferences (median score 5, n = 70) and those who do not (median score 4, n = 38; U = 1762.50, z = 2.97, r = 0.29, p = 0.003). CONCLUSIONS Virtual conferences will likely remain an integral part of neurosurgical education after the COVID-19 pandemic has abated. Across the country, residents and faculty report a preference for continued use of virtual conferencing, especially virtual case conferences and board preparation. Some traditional conferences may even be replaced with virtual conferences, in particular those that are more didactic. A new aromatic diamine, 10,10-bis [4-(4-amino-2-pyridinoxy)phenyl]-9(10H)-anthracenone (BAPPA) was synthesized from anthrone as the starting material, and the corresponding pyridine- and anthrone-containing poly(ether amide)s (PEAs) were prepared from BAPPA with various aromatic diacid chlorides via a low-temperature solution polycondensation. The resulting PEAs having number-average molecular weights (M-n) of 29,500-33,400 g/mol with the polydispersity index ranged from 1.75 to 1.84, were all amorphous, and displayed excellent thermal stability with the glass transition temperatures of 263-355 degrees C, the temperatures of 415-441 degrees C at 5% weight loss and the higher than 50% residues at 800 degrees C in nitrogen. These new PEAs were soluble in polar solvents such as N-methyl-2-pyrrolidone, N,N-dimethylacetamide and pyridine, even almost in tetrahydrofuran at room temperature and could form transparent, strong and flexible films with tensile strengths of 84-97 MPa, tensile moduli of 2.1-3.2 GPa and elongations at break of 4-13%. Also, the present PEAs exhibited high optical transparence with the UV cutoff wavelength in the 342-362 nm range, and 83-90% transmittance at 500 nm. These results indicated the introduction of anthrone and pyridine groups into the backbone could improve the solubility and transparence of polyamides.
