A new aromatic diamine, 10,10-bis [4-(4-amino-2-pyridinoxy)phenyl]-9(10H)-anthracenone (BAPPA) was synthesized from anthrone as the starting material, and the corresponding pyridine- and anthrone-containing poly(ether amide)s (PEAs) were prepared from BAPPA with various aromatic diacid chlorides via a low-temperature solution polycondensation. The resulting PEAs having number-average molecular weights (M-n) of 29,500-33,400 g/mol with the polydispersity index ranged from 1.75 to 1.84, were all amorphous, and displayed excellent thermal stability with the glass transition temperatures of 263-355 degrees C, the temperatures of 415-441 degrees C at 5% weight loss and the higher than 50% residues at 800 degrees C in nitrogen. These new PEAs were soluble in polar solvents such as N-methyl-2-pyrrolidone, N,N-dimethylacetamide and pyridine, even almost in tetrahydrofuran at room temperature and could form transparent, strong and flexible films with tensile strengths of 84-97 MPa, tensile moduli of 2.1-3.2 GPa and elongations at break of 4-13%. Also, the present PEAs exhibited high optical transparence with the UV cutoff wavelength in the 342-362 nm range, and 83-90% transmittance at 500 nm. These results indicated the introduction of anthrone and pyridine groups into the backbone could improve the solubility and transparence of polyamides.

The generation of a methyl carbon source from renewable and cheap sources is challenging. Herein, we describe a novel and an efficient route for methylation and acetylation of aza-heteroarenes using PEG-400 under O-2 and TsOH center dot H2O for the first time by tuning the reaction conditions using a different set of starting materials. The key features of the current protocol are oxidative C-O and C-C bond scission under metal-free conditions with good functional group tolerance, and a broad substrate scope. The potential applicability of the designed methodology was demonstrated for the synthesis of central nervous system (CNS) depressant and anticonvulsant drug molecules by a one-pot strategy.

In the current research, much attention is paid to heterogenized nanostructure. Herein, we report the green synthesis magnetic nanoparticles (MNPs) of cobalt ferrite by the immobilization of erbium (Er) coated with folic acid (FA) which show effective catalytic properties and recyclability. Full characterizations with field emission scanning electron microscopy (FE-SEM), transmission electron microscopy (TEM), energy-dispersive X-ray spectroscopy (EDX), X-ray atomic mapping, thermal gravimetric analysis (TGA), X-ray diffraction (XRD), vibrating sample magnetometer (VSM), inductively coupled plasma-optical emission spectrometry (ICP-OES), and Fourier-transform infrared (FT-IR) spectroscopy techniques were undertaken to uncover structural properties of the prepared magnetic catalyst. The obtained nanohybrid complexes as efficient, recyclable, and green heterogeneous systems pave the way for producing pyrano[2,3-d]pyrimidinone and dihydropyrano[3,2-c]chromenes derivatives by the one-pot three-component condensation reaction of various aldehydes, malononitrile, and hydroxycoumarin or barbituric acid in green condition. This easily prepared organometalic catalyst presents many superiorities such as operational simplicity, high yield, short reaction time, utilization of commercially available or easily accessible starting materials, eco-friendly properties, and excellent purity. Most importantly, erbium-FA-coated CoFe2O4 can be easily separated and recycled from the reaction system using an external magnetic field. The magnetically recoverable biocatalyst can be recycled and reused six times while maintaining high activities. The activity of the magnetic catalyst can be maintained at more than 80% of that of the previous cycle. This research solves the recovery problem encountered in industrial applications of biocatalysts and presents a clean and green method for preparing pyrimidinones and chromenes.

The ability of G protein-coupled receptor (GPCR) kinases (GRKs) to regulate desensitization of GPCRs has made GRK2 and GRK5 attractive targets for treating heart failure and other diseases such as cancer. Although advances have been made toward developing inhibitors that are selective for GRK2, there have been far fewer reports of GRK5 selective compounds. Herein, we describe the development of GRK5 subfamily selective inhibitors, 5 and 16d that covalently interact with a nonconserved cysteine (Cys474) unique to this subfamily. Compounds 5 and 16d feature a highly amenable pyrrolopyrimidine scaffold that affords high nanomolar to low micromolar activity that can be easily modified with Michael acceptors with various reactivities and geometries. Our work thereby establishes a new pathway toward further development of subfamily selective GRK inhibitors and establishes Cys474 as a new and useful covalent handle in GRK5 drug discovery.

Urban wildfires may generate numerous unidentified chemicals of toxicity concern. Ash samples were collected from burned residences and from an undeveloped upwind reference site, following the Tubbs fire in Sonoma County, California. The solvent extracts of ash samples were analyzed using GC- and LC-high-resolution mass spectrometry (HRMS) and using a suite of in vitro bioassays for their bioactivity toward nuclear receptors [aryl hydrocarbon receptor (AhR), estrogen receptor (ER), and androgen receptor (AR)], their influence on the expression of genetic markers of stress and inflammation [interleukin-8 (IL-8) and cyclooxygenase-2 (COX-2)], and xenobiotic metabolism [cytochrome P4501A1 (CYP1A1)]. Genetic markers (CYP1A1, IL-8, and COX-2) and AhR activity were significantly higher with wildfire samples than in solvent controls, whereas AR and ER activities generally were unaffected or reduced. The bioassay responses of samples from residential areas were not significantly different from the samples from the reference site despite differing chemical compositions. Suspect and nontarget screening was conducted to identify the chemicals responsible for elevated bioactivity using the multiple streams of HRMS data and open-source data analysis workflows. For the bioassay endpoint with the largest available database of pure compound results (AhR), nontarget features statistically related to whole sample bioassay response using Spearman’s rank-order correlation coefficients or elastic net regression were significantly more likely (by 10 and 15 times, respectively) to be known AhR agonists than the overall population of compounds tentatively identified by nontarget analysis. The findings suggest that a combination of nontarget analysis, in vitro bioassays, and statistical analysis can identify bioactive compounds in complex mixtures.

An efficient metal-free strategy for the synthesis of alpha-substituted homoallylamine derivatives has been developed via a 1,1,1,3,3,3-hexafluoro-2-propanol (HFIP)-promoted 2-aza-Cope rearrangement of aldimines, generated in situ by condensation of aldehydes with easily accessible 1,1-diphenylhomoallylamines. This reaction provides rapid access to alpha-substituted homoallylamines with excellent functional group tolerance and yields. The reaction takes place at room temperature and no chromatographic purification is required for product isolation. The synthetic utility of the current method is further demonstrated by the transformation of the obtained benzophenone ketimines into N-unprotected homoallylamines, an alpha-amino alcohol and an alpha-amino amide.
