Oxidation reactions have been extensively studied in the context of the transformations of biomass-derived furans. However, in contrast to the vast literature on utilizing the stoichiometric oxidants, such as m-CPBA and NBS, catalytic methods for the oxidative furan-recyclizations remain scarcely investigated. Given this, we report a means of manganese-catalyzed oxidations of furan with low loading, achieving the Achmatowicz rearrangement in the presence of hydrogen peroxide as an environmentally benign oxidant under mild conditions with wide functional group compatibility.

A series of novel tricyclic quinazolinone-iminosugars 1 (a-c) were synthesized from the benzyl protected sugars through three steps. Firstly, the benzyl protected sugar (aldehyde) 5 reacted with o-aminobenzamide by the iodine-induced oxidative condensation to afford the corresponding aldo-quizanolinone 6. Secondly, through the intramolecular cyclization of the unprotected OH and the amide NH in 6, the tricyclic compounds 7 and 8 were constructed by the key Mitsunobu reaction. Finally, removal of the benzyl group gave the target tricyclic quinazolinone-iminosugars 1. The protocol was effective for the preparation of the tricyclic iminosugars in satisfactory yield. Interestingly, an unusual C-2 epimerization was observed with D-mannose and D-ribose compounds under the conditions of the Mitsunobu reaction that generated the products having the trans configuration at the C-2 and C-3 positions. Unfortunately, such tricyclic quinazolinone-iminosugars showed no inhibitory effects on the tested five glycosidases.

Supported ionic liquids (SILs) have attracted rising interest and the subject of active research in the last decades due to the diversified range of applications and yet reports on ILs is still rapidly increasing. This work reports a choline sulfate ionic liquid supported on fascinating and highly stable porous graphitic carbon nitride (g-C3N4) nanosheets as an inexpensive and an environmentally friendly reusable ionic catalyst in organic synthesis typically requiring a harmful organic solvent and highly toxic adds. g-C3N4@SO3 Ch was prepared by mixing sulfonic add functionalized graphitic carbon nitride nanosheets (g-C3N4@SO3H) with choline hydroxide or via a novel approach. The introduction of a choline sulfate could significantly enlarge specific surface areas with rich reaction sites and suppressed the recombination of sheets. This work provides a new way to improve the chemical property of g-C3N4 along with the recyclability of the ionic liquid. g-C3N4@SO3 Ch (IL/g-C3N4) offers an effective, reusable, inexpensive, environmentally friendly and low-cost catalyst for the synthesis of 3,4-dihydropyrimidin-2 (IH)-ones, 2,3-dihydroquinazolin-4 (IH)-ones and bisindolylmethanes in good to excellent yields. Patterned substrates for optics, electronics, sensing, lab-on-chip technologies, bioanalytics, clinical diagnostics as well as translational and personalized medicine are typically prepared by additive substrate manufacturing including ballistic printing and microcontact printing. However, ballistic printing (e.g., ink jet and aerosol jet printing, laser-induced forward transfer) involves serial pixel-by-pixel ink deposition. Parallel additive patterning by microcontact printing is performed with solid elastomeric stamps suffering from ink depletion after a few stamp-substrate contacts. The throughput limitations of additive state-of-the art patterning thus arising may be overcome by capillary stamping -parallel additive substrate patterning without ink depletion by mesoporous silica stamps, which enable ink supply through the mesopores anytime during stamping. Thus, either arrays of substrate-bound nanoparticles or colloidal nanodispersions of detached nanoparticles are accessible. Three types of model inks are processed: 1) drug solutions, 2) solutions containing metallopolymers and block copolymers as well as 3) nanodiamond suspensions representing colloidal nanoparticle inks. Thus, aqueous colloidal nanodispersions of stamped drug nanoparticles, regularly arranged ceramic nanoparticles by post-stamping pyrolysis of stamped metallopolymeric precursor nanoparticles and regularly arranged nanodiamond nanoaggregates are obtained. Capillary stamping may overcome the throughput limitations of state-of-the-art additive substrate manufacturing while a broad range of different inks can be processed.

Based on the broad spectrum of biological activity of hydrazide-hydrazones, trifluoromethyl compounds, and clinical usage of cholinesterase inhibitors, we investigated hydrazones obtained from 4-(trifluoromethyl)benzohydrazide and various benzaldehydes or aliphatic ketones as potential inhibitors of acetylcholinesterase (AChE) and butyrylcholinesterase (BuChE). They were evaluated using Ellman’s spectrophotometric method. The hydrazide-hydrazones produced a dual inhibition of both cholinesterase enzymes with IC50 values of 46.8-137.7 mu M and 19.1-881.1 mu M for AChE and BuChE, respectively. The majority of the compounds were stronger inhibitors of AChE; four of them (2-bromobenzaldehyde, 3-(trifluoromethyl)benzaldehyde, cyclohexanone, and camphor-based 2o, 2p, 3c, and 3d, respectively) produced a balanced inhibition of the enzymes and only 2-chloro/trifluoromethyl benzylidene derivatives 2d and 2q were found to be more potent inhibitors of BuChE. 4-(Trifluoromethyl)-N’-[4-(trifluoromethyl)benzylidene]benzohydrazide 2l produced the strongest inhibition of AChE via mixed-type inhibition determined experimentally. Structure-activity relationships were identified. The compounds fit physicochemical space for targeting central nervous systems with no apparent cytotoxicity for eukaryotic cell line together. The study provides new insights into this CF3-hydrazide-hydrazone scaffold.

Herein, we report the synthesis of task-specific amino-functionalized imidazolium ionic liquid, acetate1-(2-tert-butoxycarbonylamino-ethyl)-3-methyl-3H-imidazol-1-ium; (Boc-NH-EMIM.OAc), as an efficient catalyst for the acetylation of alcohols, phenols, and amines in the presence of acetic anhydride (acetylating reagent). Remarkably, acetic anhydride in the presence of 10 mol% of catalyst (Boc-NH-EMIM.OAc) under solvent-free conditions showed excellent acetylation activity in shorter duration of time. On the basis of this, a general procedure for acetylation of alcohols, phenols, and amines has been developed. The ionic liquid (Boc-NH-EMIM.OAc) can be readily recovered and reused successfully up to four consecutive cycles without any significant loss of its catalytic activity. We have been able to show that this acetylating method has many advantages. It gives high yields, takes shorter time, and develops the possibility of benign environmental-friendly process.

The tricyclic aromatic ketone 9-anthrone and its derivatives are under basic conditions in equilibrium with their corresponding anionic forms. Unlike the neutral species, the 9-anthrolate anions can be excited by blue LED light and thus, are able to initiate a photoinduced electron transfer (PET) reaction. To demonstrate the synthetic applicability of the catalytic system, various (hetero)aryl chlorides were converted in C-C and C-Het bond-forming reactions affording the corresponding arylation products in moderate to excellent yields. The reactions proceed under very mild conditions without the need for a sacrificial electron donor. Besides 9-anthrone, other closely related derivatives were synthesised and investigated concerning their ability to catalyse demanding reductive transformations. Based on spectroscopic findings and radical trapping experiments a conceivable mechanism is proposed.

10,10-Bis[4-(4-aminophenoxy)-3-methylphenyl]-9(10H)-anthrone, as a novel aromatic diamine, was synthesized from anthrone by three-step process. A series of polyimides containing methyl substituents and 9(10H)-anthrone pendant groups was prepared from this diamine with various aromatic dianhydridesviathe poly(amic acid) (PAA) precursors and subsequent thermal or chemical imidization procedure. Inherent viscosities of PAAs are between 0.87 and 1.58 dL/g, and the corresponding polyimides have the weight-average molecular weight and number-average molecular weight in the range of 49,400-64,200 and 28,700-37,700, respectively. Most of the polyimides obtained through the chemical cyclodehydration method are soluble in NMP, DMF, DMAc, Py andm-cresol at room temperature. These polyimides have glass transition temperatures of 289-313 degrees C, 10% weight loss temperatures of 484-524 degrees C, char yields of 55-63% at 800 degrees C, and temperature at the maximum-rate degradation of 560-584 degrees C in nitrogen, respectively. The polyimide films obtained through the thermal imidization have tensile strength of 76.43-119.74 MPa, elongation at break of 6.93-12.33%, and tensile modulus of 1.91-2.38 GPa.

beta-Cyclodextrin-functionalized carbon nitride nanosheets were modified with a molecularly imprinted polymer to obtain a fluorescent probe of type MIP@beta-CD/CNNS which is shown to enable fluorometric determination of sterigmatocystin (STG). The material was characterized by transmission electron microscopy, infrared spectra, powder X-ray diffraction, X-ray photoelectron spectroscopy, and by absorption and emission spectra. The modified CNNSs have a good fluorescence quantum yield (13%), high sorption capacity for STG (86 mg center dot g(-1)), fast adsorption rate (25 min), and superior adsorption selectivity (with an imprint factor 2.56). When used as an optical probe for STG, the CNNSs act as the chromophore, while beta-CD and MIP act as the recognition groups. The blue fluorescence of MIP@beta-CD/CNNS (with excitation/emission maxima at 368/432 nm) is quenched by STG. Fluorescence drops linearly in the 0.15 to 3.1 mu M STG concentration range. The lower detection limit is 74 nM. The method was successfully applied to the determination of STG in spiked wheat extract. Conceivably, this detection scheme based on a combination of beta-CD inclusion and molecular imprinting may be extended to the detection of various other organic compounds.

PRMT4 is a type I protein arginine methyltransferase and plays important roles in various cellular processes. Overexpression of PRMT4 has been found to be involved in several types of cancers. Selective and in vivo effective PRMT4 inhibitors are needed for demonstrating PRMT4 as a promising therapeutic target. On the basis of compound 6, a weak dual PRMT4/6 inhibitor, we constructed a tetrahydroisoquinoline scaffold through a cut-and-sew scaffold hopping strategy. The subsequent SAR optimization efforts employed structure-based approach led to the identification of a novel PRMT4 inhibitor 49. Compound 49 exhibited prominently high potency and selectivity, moderate pharmacokinetic profiles, and good antitumor efficacy in acute myeloid leukemia xenograft model via oral administration, thus demonstrating this compound as a useful pharmacological tool for further target validation and drug development in cancer therapy.
