Purification of recombinant proteins is often a challenging matter because high purity and high recovery are desired. If the expressed recombinant protein is also in a complex matrix, such as from the silkworm expression system, purification becomes more challenging. Even if purification from the silkworm expression system is troublesome, it benefits from a high capacity for the production of recombinant proteins. In this study, magnetic nanoparticles (MNPs) were investigated as a suitable tool for the purification of proteins from the complex matrix of the silkworm fat body. The MNPs were modified with nickel so that they have an affinity for His-tagged proteins, as the MNP purification protocol itself does not need special equipment except for a magnet. Among the three different kinds of investigated MNPs, MNPs with sizes of 100 nm to 200 nm and approximately 20 nm-thick nickel shells were the most suitable for our purpose. With them, the total protein amount was reduced by up to at least approximately 77.7%, with a protein recovery of around 50.8% from the silkworm fat body. The minimum binding capacity was estimated to be 83.3 mu g protein/mg MNP. Therefore, these MNPs are a promising tool as a purification pretreatment of complex sample matrices.

A 2,2 ‘-Bipyridine-based ionic compound named 2,2 ‘-bipyridinium dihydrogen phosphate was synthesized by addition of phosphoric acid to a solution of 2,2 ‘-Bipyridine in dichloromethane. After the characterization using FT-IR, mass, H-1, C-13 and P-31 NMR techniques, it was used as a Bronsted dicationic acidic catalyst for the promotion of the synthesis of 2-arylidene malononitrile and 5-arylidene barbituric acid derivatives via Knoevenagel condensation reaction in water. Graphene oxide was covalently modified with p-phenylenediamine via a diazonium reaction. The resulting material was employed as a sorbent for the solid-phase extraction of six phenylurea herbicides (metoxuron, monuron, chlortoluron, isoproturon, monolinuron, and buturon) from environmental water and lettuce leafs. Some key factors that influence the extraction efficiency were studied, including sample loading rate, sample pH, and desorption conditions. Following desorption with acetonitrile, the analytes were quantified by HPLC with UV detection. Under optimized conditions, response to phenylurea herbicides is linear in the 2.0-100ngmL(-1) concentration range for water samples, and 5.0-100ngg(-1) for leaf lettuces. The limits of detection are 0.10-0.25ngmL(-1) for water samples, and 1.5-2.5ngg(-1) for leaf lettuces. The sorbent was also applied to the preconcentration of organic compounds including nitroimidazoles, chlorophenols, phenylurea insecticides and phthalates. This shows that this sorbent has a large potential for the enrichment of organic pollutants.

Organic pollutants associated with diesel exhaust particles (DEP), such as polycyclic aromatic hydrocarbons (PAHs) and their derivatives, may negatively impact human health. However, a comprehensive overview of their effects on endocrine nuclear receptor activities is still missing. Here, we evaluated the effects of extracts and chromatographic fractions (fractionated according to increasing polarity) of two standard reference materials derived from distinct types of diesel engines (SRM 2975, SRM 1650b), on activation of androgen receptor (AR), estrogen receptor alpha (ER alpha), peroxisome proliferator-activated receptor gamma (PPAR gamma), glucocorticoid receptor (GR) and thyroid receptor alpha (TR alpha), using human cell-based reporter gene assays. Neither DEP standard modulated AR or GR activities. Crude extracts and fractions of SRM 1650b and SRM 2975 suppressed ER alpha-mediated activity in the ER-CALUX (TM) assay; however, this effect could be partly linked to their cytotoxicity in this cell line. We observed that only SRM 2975 extract and its fractions were partial PPAR gamma inducers, while SRM 1650b extract was not active towards this receptor. Importantly, we found that both extracts and polar fractions of SRM activated TR alpha and significantly potentiated the activity of endogenous TR alpha ligand, triiodothyronine. Based on a detailed chemical analysis of both extracts and their polar fractions, we identified several oxygenated PAH derivatives, that were present at relatively high levels in the analyzed DEP standards, including 3-nitrobenzanthrone (3-NBA), anthracene-9,10-dione, phenanthrene-9,10-dione. 9H-fluoren-9-one or benzo[a] anthracene-7,12-dione, to activate TR alpha activity. Nevertheless, these compounds provided only a minor contribution to the overall TR alpha activity identified in polar fractions. This suggests that yet unidentified polar polyaromatic compounds associated with DEP may, apart from their known impact on the aryl hydrocarbon receptor or steroid signaling, deregulate activities of additional nuclear receptors, in particular of TR alpha. A new epoxy monomer, namely, tetraglycidyl-1,2-aminobenzamide (ER), was synthesized by condensation of the amines with epichlorohydrin in a basic medium. The obtained epoxy monomer was characterized by FT-IR and H-1 NMR spectroscopy. Rheological properties of this monomer were determined using an advanced rheometer. Subsequently, the synthesized ER monomer was investigated as corrosion inhibitor for carbon steel in 1 M HCl solution. The adsorption properties of ER were analyzed by electrochemical, surface investigation and theoretical computational studies using DFT and molecular dynamics (MD). Results showed a high dependence of the viscosity of ER on temperature and concentration, and also, that ER has better inhibition performance. A good agreement between the results derived from computational (MD and DFT) and experimental methods was observed. The thermodynamic parameters, along with the kinetic parameters, showed that the adsorption of ER molecules onto carbon steel surface obeyed the Langmuir isotherm model, and the adsorption at metal-electrolyte interfaces involved both chemical and physical adsorption, but predominantly chemisorption mechanism.

Flexible fiber-type solid-state supercapacitors (FSCs) are demanded for energy storage of wearable soft electronics. A flexible carbon fiber (CF) with high electrical conductivity and good energy storage ability can act as the current collector and the active material simultaneously. Improving the energy storage ability of the CF without depositing active materials is highly promising to avoid active materials from peeling off, especially under bending conditions. In this study, the CF is treated with acid mixtures containing different HNO3 to H2SO4 ratios. The roughened surface and the production of graphene quantum dots and functional groups are achieved for the acid-treated CF, especially for the CF treated with the acid mixtures. The largely enhanced specific capacitance (C-L) is first obtained for the optimized CF electrode (641.08 mF/cm at 5 mV/s) treated with the acid mixture containing the HNO3 to H2SO4 ratio of 1 to 3. The FSC also shows a maximum energy density of 16.57 mu W h/cm at the power density of 62.50 mu W/cm, as well as a C-L retention of 77% and Coulombic efficiency of 100% in 5000 times of a charge/discharge cycling process. Excellent flexibility is also attained for the FSC with no shape distortion of cyclic voltammetry curves measured under the bending angle of 180 degrees and with 1000 times of repeated bending. This work proposes a simple acid treatment to largely enhance the energy storage ability of pure CF electrodes. The bifunctional substance with current collecting and energy storing abilities is expected to be well developed in the future for applying on numerous efficient flexible energy storage devices.

Background: Colorectal familial adenomatous polyposis (FAP) adenomas exhibit a uniform pathogenetic basis caused by a germline mutation in the adenomatous polyposis gene (APC), but the molecular changes leading to their development are incompletely understood. However, dysregulated apoptosis is known to substantially affect the development of colonic adenomas. One of the key regulatory proteins involved in apoptosis is apoptosis repressor with caspase recruitment domain (ARC). Methods: The expression of nuclear and cytoplasmic ARC in 212 adenomas from 80 patients was analyzed by immunohistochemistry. We also compared expression levels of ARC with the expression levels of p53, Bcl-2, COX-2, and MMR proteins. Statistical analyses were performed by Spearman’s rank correlation and linear regression test. Results: ARC was overexpressed in the nuclei and cytoplasm of most FAP adenomas investigated. Cytoplasmic ARC staining was moderately stronger (score 2) in 49.1% (n = 104/212) and substantially stronger (score 3) in 32.5% (n = 69/212) of adenomas compared to non-tumorous colorectal mucosa. In 18.4% (n = 39/212) of adenomas, cytoplasmic ARC staining was equivalent to that in non-tumorous mucosa. Nuclear expression of ARC in over 75% of cells was present in 30.7% (n = 65/212) of investigated adenomas, and nuclear expression in 10-75% of cells was detected in 62.7% (n = 133/212). ARC expression in under 10% of nuclei was found in 6.6% (n = 14/212) of adenomas. The correlation between nuclear ARC expression and cytoplasmic ARC expression was highly significant (p = 0.001). Moreover, nuclear ARC expression correlated positively with overexpression of Bcl-2, COX-2 p53 and beta-catenin. Cytoplasmic ARC also correlated with overexpression of Bcl-2. Sporadic MMR deficiency was detected in very few FAP adenomas and showed no correlation with nuclear or cytoplasmic ARC. Conclusions: Our results demonstrated that both cytoplasmic and nuclear ARC are overexpressed in FAP adenomas, thus in a homogenous collective. The highly significant correlation between nuclear ARC and nuclear beta-catenin suggested that ARC might be regulated by beta-catenin in FAP adenomas. Because of its further correlations with p53, Bcl-2, and COX-2, nuclear ARC might play a substantial role not only in carcinomas but also in precursor lesions.

Group 6 alkynyl Fischer carbene complexes behave as electrophiles with highly reactive masked dienes such as anthrone and 3-hydroxy-2-pyrone, which also can react as bidentate nucleophiles. Both Cr-0 and W-0 alkynyl carbene complexes require tertiary amines to promote the formation of 1,4-C-addition products, but a marked dependence of the reactivity on the metal was observed. Although W-0 complexes form exclusively the 1,4-C-adducts in the presence of a catalytic amount of base, Cr-0 complexes lead to new anthrone-derived bis-carbene complexes, obtained by a sequential 1,4-C- and 1,4-O-addition process with high diastereoselectivity and only in the presence of an excess of base. DFT computational studies show that the gap between the LUMO of the complex and the HOMO of the anthrone determines the differences in reactivity and that the stability of the intermediate metal allenyls formed after the initial 1,4-addition is decisive for the outcome of the reaction.
