A new regioselective 3,4-difunctionalization of 3-chloropyridines via 3,4-pyridyne intermediates is reported. Regioselective lithiation of 3-chloro-2-ethoxypyridine and a related 2-thio-derivative followed by treatment with aryl- and alkylmagnesium halides as well as magnesium thiolates at -78 degrees C produced 3,4-pyridynes during heating to 75 degrees C. Regioselective addition of the Grignard moiety in position 4 followed by an electrophilic quench in position 3 led to various 2,3,4-trisubstituted pyridines. This method was adapted into a continuous flow set-up. As an application, we have prepared a key intermediate for (+/-)-paroxetine.

We report aggregation induced emission (AIE) driven polysaccharide polymersomes as fluorescence resonance energy transfer (FRET) nanoprobes to study their intracellular enzyme-responsive delivery by real-time live-cell confocal microscopy bio-imaging techniques. An AIE active tetraphenylethylene (TPE) optical chromophore and plant-based vesicular directing hydrophobic unit were grafted on clinically relevant polysaccharide-dextran via enzyme-cleavable aliphatic ester chemical linkages. The TPE-tagged dextran self-assembled as 180 +/- 20 nm blue-luminescent polymersomes in aqueous medium and exhibited excellent encapsulation capabilities for water soluble Rose Bengal (RB) and water insoluble Nile red (NR) fluorophores. The selective photoexcitation of the TPE chromophore enabled the FRET process between the TPE donor and RB (or NR) acceptor molecule in <50 angstrom Forster distance afforded by the polymersome. The FRET probe was very stable under extracellular conditions and it exclusively underwent lysosomal esterase enzymatic biodegradation at the intracellular compartments to release RB. The enzyme-trigger enabled the FRET probe to function as an extracellular turn-ON -> intracellular turn-Off red-fluorescent signal (Probe-1). In this process, the AIE self-emission was also simultaneously restored on the TPE chromophore (blue-luminescent, Probe-2) followed by the isolation of donor and acceptor in the cytosol. As a result, this new design enabled the visualization of real-time enzyme-responsive delivery by monitoring the dual fluorescent signals from both the polymer host (blue) and encapsulated guest (red) in a single nano-platform. In vitro cytotoxicity studies established that the polymersome probe was non-toxic to cells up to 300 mu g mL(-1). Lyso-tracker staining experiments supported the FRET probe internalization in the lysosomal compartments for enzymatic-biodegradation. Live cell confocal microscopy with selective photo-excitation was used to directly monitor the enzyme-responsive FRET action in human breast cancer MCF 7 and wild-type mouse embryonic fibroblast cell lines (WT-MEFs). It was found that the tailor-made polymersome FRET probe was efficient to deliver the loaded cargo in <3 h in live cells which predicts the usefulness of the probe in biomedical research. Herein is reported a substantially improved synthesis of 2-substituted benzimidazoles by condensation of 1,2-diaminoarenes and aldehydes using methanol as the reaction medium. The developed method afforded moderate to excellent yields (33-96%) at ambient temperature, displays high functional group tolerance, is conducted open to air, and requires only one minute reaction time under catalyst- and additive-free conditions. Moreover, the efficient protocol permits scale-up to multi-gram scale synthesis of benzimidazoles and will become a method of choice when constructing such heterocyclic systems.

Kinetics of thermostimulated luminescence (TSL) of poly(diphenylenephthalide) (PDPh) films has been studied using mathematical modelling and DFT approximation. TSL of PDPh-film was considered to be arising from irradiative deactivation of diphenylene (DP) fragment of polymer chain in excited state formed upon recombination of the spatial separated ion-radical pairs generated by photoexcitation. Two parallel monomolecular transformations leading to excited DP have been revealed: the first path is characterized by the activation parameters of the kinetic rate constant k1 which equal to Al ti 105 sec 1 and Eat ti 70 kJ mol(-1). This path may be interpreted in terms of mechanical relaxation theory by segmental polymer mobility as electron transfer (ET) from phthalide (Ph) anion-to DP cation-radical. The second channel is supposed to be consist of two consecutive ET from triarylmethyl anion-radical to phthalide neutral (A2 ti 1 sec 1, Ea2 ti 20 kJ mol(-1)), and then to DP cation-radical (A3 N 104 sec 1, E,3 N 40 kJ mol(-1)) of the polymer chain indicating y- and fl-relaxation, respectively. The revealed mechanistic steps and electron retention efficiency estimated in B97-2/6-311 + G(d,p) approximation for the most probable anion-radicals may point out the key role of domino quantum tunneling in transfer and recombination of charges in PDPh films.

The application of carboxy-MIDA-boronate (MIDA=N-methyliminodiacetic acid) as an in situ CO surrogate for various palladium-catalyzed transformations is described. Carboxy-MIDA-boronate was previously shown to be a bench-stable boron-containing building block for the synthesis of borylated heterocycles. The present study demonstrates that, in addition to its utility as a precursor to heterocycle synthesis, carboxy-MIDA-boronate is an excellent in situ CO surrogate that is tolerant of reactive functionalities such as amines, alcohols, and carbon-based nucleophiles. Its wide functional-group compatibility is highlighted in the palladium-catalyzed aminocarbonylation, alkoxycarbonylation, carbonylative Sonogashira coupling, and carbonylative Suzuki-Miyaura coupling of aryl halides. A variety of amides, esters, (hetero)aromatic ynones, and bis(hetero)aryl ketones were synthesized in good-to-excellent yields in a one-pot fashion.

A series of organic host-guest materials with multifunctional luminescence were constructed. Four isoquinoline derivatives were used as the guests, and benzophenone was used as the host. The doped system exhibited excellent dual emission with cyan fluorescence and orange-yellow room-temperature phosphorescence, and the dual emission could be combined into almost pure white-light emission. Importantly, the relative intensity of the fluorescence-phosphorescence could be adjusted by changing the excitation wavelength, with the phosphorescence intensity being significantly higher than the fluorescence intensity under shorter excitation wavelengths and vice versa under longer excitation wavelengths. Therefore, three-color emission switching among cyan, white, and orange could be achieved by simply adjusting the excitation wavelength. The results of experimental and theoretical calculations indicated that the excitation-dependent emission colors were caused by different transfer paths for excitons under different excitation wavelengths. These materials with multifunctional luminescence could be used as writable inks for advanced anticounterfeiting.

Houttuynia essential oil (HEO) has excellent antiviral, anti-inflammatory, and other pharmacological effects, but the lack of effective analytical methods to quantify HEO in plasma has hindered its better clinical monitoring. Houttuynine (Hou) is one of the main active ingredients and quality control substances of HEO, so the pharmacokinetic study of HEO could be conducted by determining Hou blood concentration. Hou is active and not stable in plasma, which makes its blood concentration difficult to measure. In this work, a novel liquid chromatography tandem mass spectrometry (LC-MS/MS) method for Hou determination in rat blood was established that involves Hou being derivatized with 2, 4-dinitrophenylhydrazine to form a stable compound to prevent degradation. Herein, p-Tolualdehyde-2,4-dinitrophenylphenylhydrazone was selected as an internal standard substance and the LC-MS/MS method was evaluated for selectivity, precision, accuracy, calibration limit, matrix effect, recovery, and stability. Good linearity (r(2) = 0.998) was reached in the range of 2-2000 ng/mL, and the lower limit of quantification of Hou was determined to be 2 ng/mL. The mean intra-assay accuracy ranged from 77.7% to 115.6%, whereas the intra-assay precision (relative standard deviation, RSD) was below 11.42%. The matrix effect value for Hou in rat plasma was greater than 75%, and for the internal standard (IS) it was 104.56% +/- 3.62%. The extraction recovery of Hou were no less than 90%, and for the IS it was 96.50% +/- 4.68%. Our method is sensitive and reliable and has been successfully applied to the pharmacokinetic analysis of Hou in rats given HEO via gavage and injection.

A novel emulsified photoinitiator (named as OP10-IPDI-2959) was prepared by the reaction of isophorone diisocyanate (IPDI) with 2-hydroxy-4?-(2-hydroxyethoxy)-2-methylpropiophenone (Ir-2959) and nonylphenol polyoxyethylene ether (OP-10). The structure of OP10-IPDI-2959 was characterized by FT-IR, 1H NMR, UV?vis and high-resolution mass spectrometry. The photochemical behavior of OP10-IPDI-2959 was investigated through the photolysis and polymerization of neopentyl glycol diacrylate (NPGDA), mixture of epoxy acrylate resin and 1,6-hexanediol diacrylate (HDDA). Under irradiation of UV light, the OP10-IPDI-2959 has similar photochemical properties with the Ir-2959. Combined with benzophenone, the photoinitiated rate of OP10-IPDI-2959 is higher than that of Ir-2959. The polyoxyethylene fragment of OP10-IPDI-2959 is not only an alternative to toxic amine as hydrogen donor, but also a hydrophilic unit for interface between oil and water. The OP10-IPDI-2959 can effectively emulsify the epoxy acrylate resin and 1,6-hexanediol diacrylate mixture to form low viscosity emulsion. Compared with Ir-2959, OP10-IPDI-2959 has good photoinitiation activity, lower migration and good water emulsification performance.

Solid-contact ion-selective electrodes (SC-ISEs) have shown great potential for routine and portable ion detection. The introduction of nanomaterials as ion-to-electron transducers and the adoption of different performance-enhancement strategies have significantly promoted the development of SC-ISEs. Herein, new solid-contact ion-selective electrodes, along with the implementation of multiwalled carbon nanotubes (MWCNTs) as ion-to-electron transducers and potassium tetrakis (p-chlorophenyl) borate (KTpClB) as lipophilic ionic additives, were presented for the detection of isoproturon (IPU) and diuron (DU) herbicides. Molecularly imprinted polymers (MIPs), with special molecule recognition properties for isoproturon (IPU) and diuron (DU), were prepared, characterized, and introduced as sensory recognition materials in the presented electrodes. Sensors revealed a near-Nernstian response for both isoproturon (IPU) and diuron (DU) with slopes of 53.1 +/- 1.2 (r(2) = 0.997) and 57.2 +/- 0.3 (r(2) = 0.998) over the linear ranges of 2.2 x 10(-6)-1.0 x 10(-3) M and 3.2 x 10(-6)-1.0 x 10(-3) M with detection limits of 8.3 x 10(-7) and 1.4 x 10(-6) M, respectively. The response time of the presented sensors was found to be <5 s and the lifetime was at least eight weeks. The sensors exhibited good selectivity towards isoproturon (IPU) and diuron (DU) in comparison with some other herbicides, alkali, alkaline earth, and heavy metal ions. The chiral building block (R)-(+)-2,2 ‘-diamino-1,1 ‘-binaphthyl, (R)-BINAM, which is often used as backbone in privileged enantioselective catalysts, was converted to a series of N-substituted proligands (R)1-H-2 (R = CH(2)tBu, C(H)Ph-2, PPh2, dibenzosuberane, 8-quinoline). After double deprotonation with strong Mg or Ca bases, a series of alkaline earth (Ae) metal catalysts (R)1-Ae.(THF)(n) was obtained. Crystal structures of these C-2-symmetric catalysts have been analyzed by quadrant models which show that the ligands with C(H)Ph-2, dibenzosuberane and 8-quinoline substituents should give the best steric discrimination for the enantioselective intramolecular alkene hydroamination (IAH) of the aminoalkenes H2C = CHCH2CR ‘ 2CH2NH2 (CR ‘(2) = CPh2, CCy or CMe2). The dianionic (R)1(2-) ligand in (R)1-Ae center dot(THF)(n) functions as reagent that deprotonates the aminoalkene substrate, while the monoanionic ((R)1-H)(-) ligand formed in this reaction functions as a chiral spectator ligand that controls the enantioselectivity of the ring closure reaction. Depending on the substituent R in the BINAM ligand, full cyclization of aminoalkenes to chiral pyrrolidine products as fast as 5 minutes was observed. Product analysis furnished enantioselectivities up to 57% ee, which marks the highest enantioselectivity reported for Ca catalyzed IAH. Higher selectivities are impeded by double protonation of the (R)1(2-) ligand leading to complete loss of chiral information in the catalytically active species.
