Solid-contact ion-selective electrodes (SC-ISEs) have shown great potential for routine and portable ion detection. The introduction of nanomaterials as ion-to-electron transducers and the adoption of different performance-enhancement strategies have significantly promoted the development of SC-ISEs. Herein, new solid-contact ion-selective electrodes, along with the implementation of multiwalled carbon nanotubes (MWCNTs) as ion-to-electron transducers and potassium tetrakis (p-chlorophenyl) borate (KTpClB) as lipophilic ionic additives, were presented for the detection of isoproturon (IPU) and diuron (DU) herbicides. Molecularly imprinted polymers (MIPs), with special molecule recognition properties for isoproturon (IPU) and diuron (DU), were prepared, characterized, and introduced as sensory recognition materials in the presented electrodes. Sensors revealed a near-Nernstian response for both isoproturon (IPU) and diuron (DU) with slopes of 53.1 +/- 1.2 (r(2) = 0.997) and 57.2 +/- 0.3 (r(2) = 0.998) over the linear ranges of 2.2 x 10(-6)-1.0 x 10(-3) M and 3.2 x 10(-6)-1.0 x 10(-3) M with detection limits of 8.3 x 10(-7) and 1.4 x 10(-6) M, respectively. The response time of the presented sensors was found to be <5 s and the lifetime was at least eight weeks. The sensors exhibited good selectivity towards isoproturon (IPU) and diuron (DU) in comparison with some other herbicides, alkali, alkaline earth, and heavy metal ions. In recent years, supercritical fluid chromatography (SFC) has become a powerful tool in modern analytical chemistry, and the diversity of stationary phases in SFC promotes phenyl-type phases to confront with a significant resurgence of interest. In this paper, a series of phenyl-type stationary phases with different substituted benzenes involving N-propylbenzamide (PB), 4-fluoro-N-propylbenzamide (PB-F), and 4-ethyl-N-propylbenzamide (PB-ET) were synthesized. Retention mechanism of these phases in SFC was investigated using a linear solvation energy relationship (LSER) model. The phenyl-type stationary phases with all positive parameters can provide all types of interaction, typically involving hydrogen bonding, dipole-dipole and dispersive interactions. The different benzene’s substituents of the stationary phases mainly affected their hydrogen bonding and dipole-dipole interactions, which could be reflected by the angle between the solvation vectors to some extent. The k-k plot showed that the selectivity difference of phenyl-type stationary phases was closely related to the type of solute. Thus, based on twenty-five natural phenolic compounds, two systems with high orthogonality (63.49%) were constructed using three columns, namely phenyl column (PHE) x PB-F and PB x PB-F. Finally, after investigating the influence of chromatographic conditions, ten flavonoids could be separated by using PB, PB-F and PHE columns in SFC. (C) 2019 Published by Elsevier B.V.

Biodegradation of the phenylurea herbicide linuron appears a specialization within a specific Glade of the Variovorax genus. The linuron catabolic ability is likely acquired by horizontal gene transfer but the mechanisms involved are not known. The full-genome sequences of six linuron-degrading Variovorax strains isolated from geographically distant locations were analyzed to acquire insight into the mechanisms of genetic adaptation toward linuron metabolism. Whole-genome sequence analysis confirmed the phylogenetic position of the linuron degraders in a separate Glade within Variovorax and indicated that they unlikely originate from a common ancestral linuron degrader. The linuron degraders differentiated from Variovorax strains that do not degrade linuron by the presence of multiple plasmids of 20-839 kb, including plasmids of unknown plasmid groups. The linuron catabolic gene clusters showed 1) high conservation and synteny and 2) strain-dependent distribution among the different plasmids. Most of them were bordered by IS1071 elements forming composite transposon structures, often in a multimeric array configuration, appointing IS1071 as a key element in the recruitment of linuron catabolic genes in Variovorax. Most of the strains carried at least one (catabolic) broad host range plasmid that might have been a second instrument for catabolic gene acquisition. We conclude that Glade 1 Variovorax strains, despite their different geographical origin, made use of a limited genetic repertoire regarding both catabolic functions and vehicles to acquire linuron biodegradation.

This study evaluated the remobilization, mineralization, and metabolism of herbicide-bound residues in soils amended with various types of sugarcane waste. Soil with bound residues of three herbicides and fresh soil samples were added to the biometric flasks, followed by the addition of vinasse, filter cake, or sugarcane straw in order to reactivate the microbial activity. In sandy loam soil, higher mineralization was observed where filter cake was added, and the maximum mineralized percentage was 7.7, 46.7, and 8.1% of diuron-, hexazinone-, and metribuzin-bound residues, respectively. Conversely, this soil presented a greater percentage of re-extractable hexazinone and metribuzin residues when vinasse was added. Among the examined herbicides, a higher percentage of bound metribuzin residues remained in the soil (57.5-75.6%). It was possible to identify both metabolites and parent compounds in the re-extracted residues, implying either species could bind to the soil. Therefore, this study has shown that bound residues of three herbicides and their metabolites can become bioavailable, and mineralized or returned to the soil solution, which could adversely affect subsequent crops or non-target organisms. Hence, the remobilization of bound residues must be taken into account when assessing the environmental risk of herbicides in soils in registration processes.

Objective: Androgen metabolism is a key component in therapeutic resistance to androgen deprivation therapy (ADT). This study aimed to reveal the significance of genetic polymorphisms in genes involved in androgen metabolism, including CYP17A1, AKR1C3, and HSD17B, on serum testosterone levels during ADT, as well as the prognosis of men undergoing ADT for metastatic prostate cancer (CaP). Materials and methods: This study included 104 Japanese patients with metastatic CaP, for whom serum testosterone data during ADT were available for 80 patients. The association of CYP17A1 (rs743572), AKR1C3 (rs12529), HSD17B1 (rs605059), HSD17B3 (rs2066479), and HSD17B4 (rs7737181) with serum testosterone levels during ADT and prognosis (progression-free survival and overall survival) was examined. Enzymatic activity in AKR1C3 H5Q was examined using recombinant protein. Results: Homozygous wild-type (GG allele; median [interquartile range], 12.0 ng/ml [8.0-19.0 ng/ml]) AKR1C3 rs12529 was associated with higher serum testosterone levels during ADT compared with variant-type (GC/CC alleles; median [interquartile range], 9.0 ng/ml [6.4-10.8 ng/ml]). Consistently, variant-type (GC/CC alleles) AKR1C3 rs12529 showed significantly lower risk of progression (hazard ratio [95% confidence interval], 0.47 [0.24-0.96], P = 0.039) compared with homozygous wild-type (GG allele) on multivariate analysis. Meanwhile, other genetic variations were associated with neither serum testosterone during ADT nor prognosis. Enzyme activity of wild-type AKR1C3 was comparable to the H5Q mutant. Conclusions: Taken together, this study demonstrated that AKR1C3 polymorphism, which was associated with serum testosterone levels during ADT, may be a prognostic factor of the progression to castration-resistant prostate cancer in Japanese men with metastatic CaP. The EPHA2 tyrosine kinase receptor is implicated in tumor progression and targeted therapies resistance. We evaluated EPHA2 as a potential resistance marker to the antiepidermal growth factor receptor (EGFR) monoclonal antibody cetuximab in colorectal cancer. We studied activation of EPHA2 in a panel of human colorectal cancer cell lines sensitive or resistant to anti-EGFR drugs. The in vitro and in vivo effects of ALW-II-41-27 (an EPHA2 inhibitor) and/or cetuximab treatment were tested. Fonnalin-fixed paraffin-embedded tumor specimens from 82 RAS wild-type (WV) metastatic colorectal cancer patients treated with FOLFIRI + cetuximab as first-line therapy in the CAPRI-COIM trial were assessed for EPHA2 expression by immunohistochemistry and correlated with treatment efficacy. EPHA2 was differentially activated in colorectal cancer cell lines. Combined treatment with ALW-II-41-27 plus cetuximab reverted primary and acquired resistance to cetuximab, causing cell growth inhibition, inducing apoptosis and cell-cycle G1-G2 arrest. In tumor xenograft models, upon progression to cetuximab, ALW-II-41-27 addition significantly inhibited tumor growth. EPHA2 protein expression was detected in 55 of 82 tumor samples, frequently expressed in less-differentiated and left-sided tumors. High levels of EPHA2 significantly correlated with worse progression-free survival [8.6 months; confidence interval (CI) 95%, 6.4-10.8; vs. 12.3 months; CI 95%, 10.4-14,2; P = 0.03] and with increased progression rate (29% vs. 9%, P = 0.02). A specific EPHA2 inhibitor reverts in vitro and in vivo primary and acquired resistance to anti-ECFR therapy. EPHA2 levels are significantly associated with worse outcome in patients treated with FOLFIRI + cetuximab. These results highlight EPHA2 as a potential therapeutic target in metastatic colorectal cancer.

This study reports an innovative pathway for successfully synthesizing composite panels using various waste input. For this purpose, seven types of powder from waste or widely available filler i.e. Quartz off-cut, sand, waste seashell, dolomite, limestone aggregates, concrete waste and limestone dust were used. The study aims to assess the effectiveness and mechanical properties of the various waste powder in the production of powder-resin composites. The panels were then compared with the novel polymeric glass composite (PGC) in the previous study (Heriyanto et al., 2018). Following the same procedure in PGC, the filler was individually grounded to 64-108 mu m and chemically treated with amino silane coupling agent (CA). The powder filler (untreated or treated) is then mixed with the resin binder with a ratio of 80/20 respectively, followed by hot pressing the mixture at the pressure of 550 bar at 65 degrees C for 1 h. The final composite slab is then further cut for mechanical testing. It was found in the study that when CA was not added, surface roughness of the powder particles affected the flexural strength of the final panel significantly. High surface roughness particles such as in Quartz, sand and seashell adhere effectively with the resin binder which led to higher strength. On the contrary, other factors like smooth particle morphology in glass, dolomite and limestone as well as porous structure in concrete and clump of very fine powder in limestone dust degrade the strength of the final panels. With CA addition, adhesion between resin and powder filler were improved significantly. Flexural strength after the CA treatment was found to be much affected by particle characteristics. Silica-based panels i.e. quartz, sand and glass which consist of high strength and hardness of silica particles perform better compared to that of calcium carbonate-based panels. Compression strength, toughness, stiffness, scratch resistance, density and water absorption were also reported in this study. The properties of all the treated panels are found to be comparable, or if not, much better than natural stones. These new approaches of using waste filler in powder – resin based composites can be a new alternative to produce green materials that deliver economic and environmental benefits. A photoinduced synthesis of S-aryl thiosulfonates through a three-component reaction of aryldiazonium tetrafluoroborates, sodium metabisulfite, and thiourea is achieved. The reaction scope generality with a range of aryldiazonium tetrafluoroborates is demonstrated. In this transformation, a radical coupling pathway is proposed with the insertion of sulfur dioxide in the presence of a photocatalyst under visible light irradiation. The convenient preparation of N2-unprotected five-membered cyclic guanidines was achieved through a cascade [3 + 2] cycloaddition between organo-cyanamides and alpha-haloamides under mild conditions in good to excellent yields (up to 99%). Graphene oxide was covalently modified with p-phenylenediamine via a diazonium reaction. The resulting material was employed as a sorbent for the solid-phase extraction of six phenylurea herbicides (metoxuron, monuron, chlortoluron, isoproturon, monolinuron, and buturon) from environmental water and lettuce leafs. Some key factors that influence the extraction efficiency were studied, including sample loading rate, sample pH, and desorption conditions. Following desorption with acetonitrile, the analytes were quantified by HPLC with UV detection. Under optimized conditions, response to phenylurea herbicides is linear in the 2.0-100ngmL(-1) concentration range for water samples, and 5.0-100ngg(-1) for leaf lettuces. The limits of detection are 0.10-0.25ngmL(-1) for water samples, and 1.5-2.5ngg(-1) for leaf lettuces. The sorbent was also applied to the preconcentration of organic compounds including nitroimidazoles, chlorophenols, phenylurea insecticides and phthalates. This shows that this sorbent has a large potential for the enrichment of organic pollutants.
