12-Deoxyphorbol-13-phenylacetate (DPP), found in Euphorbia resinifera, is a prototype of a new class of phorbol derivatives that function as protein kinase C activators with potent anti-tumor-promoting activity. To better understand the biosynthesis of DPP in E. resinifera and develop methods for its production, we used next-generation sequencing technologies to build a transcriptome dataset. We obtained a total of approximately 27,102,078 clean reads that were assembled into 90,448 unigenes. Of those, 129 and 59 unigenes were identified in the Terpenoid Backbone Biosynthesis (TBB) (KEGG map00900) and the Diterpenoid Biosynthesis (DB) (KEGG map00904) pathways, respectively. We investigated the expression patterns of 23 DPP biosynthesis-related genes and validated the results by quantitative real time PCR. The results showed that CMK (4-diphosphocytidyl-2-C-methyl-o-erythritol kinase), MECS (2-C-methyl-D-erythritol 2,4-cyclodiphosphate synthase), HDS (1-hydroxy-2-methyl-2-(E)-butenyl 4-diphosphate synthase), and IPI (Isopentenyl-diphosphate Delta-isomerase) of the methylerythritol 4-phosphate (MEP) pathway had a higher expression compared to other genes of TBB, whereas the expression levels of genes involved in the mevalonic acid (MVA) pathway were relatively lower and not significantly different from each other. Furthermore, the expression of the 23 genes in root, stem and young stem differed, and the statistical results indicated that HMGS (Hydroxymethylglutaryl-CoA synthase), MVK (mevalonic acid), MECT (2-C-methyl-D-erythritol 4-phosphate cytidylyltransferase), MECS, HDS and IPI in the TBB pathway as well as KS in the DB pathway were significantly and positively correlated with the CS (casbene synthase) gene. These results suggested that both cytosol-localized MVA and plastid-localized MEP pathways may have a significant contribution in DPP accumulation, with MEP pathway playing a dominant role. To the best of our knowledge, this study is the first transcriptome analysis of E. resinifera and the results reported here will aid the identification of functional genes and better understanding of DPP biosynthesis.

Several ethylenedioxy-bridged bisarenes with a variety of type and number of aryl groups were synthesized to study non-covalent dispersion-driven inter- and intramolecular aryl-aryl interactions in the solid state and gas phase. DFT calculations with and without dispersion correction show larger interacting aromatic groups increase the stabilization energy of folded conformers and decrease the intermolecular centroid-centroid distance. Gas electron diffraction experiments were performed exemplarily for 1-(pentafluorophenoxy)-2-(phenoxy)ethane. A new procedure for structure refinement was developed to deal with the conformational complexity of such molecules. The results are an experimental confirmation of the existence of folded conformations of this molecule with short intramolecular aryl-aryl distances in the gas phase. Solid-state structures are dominated by stretched structures without intramolecular aryl-aryl interactions but interactions with neighboring molecules.

Chiral hyperbranched polymers (HBP) containing cinchona alkaloids were synthesized using a Mizoroki-Heck (MH) coupling polymerization reaction between a cinchona squaramide dimer and tri- or tetrasubstituted aromatic iodides. This is a new type of polymeric chiral organocatalyst. We found that the as-obtained chiral HBPs show excellent catalytic activity in the asymmetric Michael reaction. Almost perfect enantioselectivity (>99% ee) was achieved in the reaction of beta-ketoester and trans-beta-nitrostyrene. Chiral polyureas of cinchona alkaloids were synthesized via repetitive Mizoroki-Heck (MH) coupling reaction. The Pd-catalyzed polycondensation of cinchona urea dimers6and aromatic diiodide7afforded the chiral polyureas (P1-P4). The catalytic activity of the chiral polymers was subsequently investigated. The asymmetric Michael addition of ketoesters to nitroolefins was successfully catalyzed by the polymeric organocatalysts (P1-P4) to give the corresponding Michael adducts with high catalytic activities and excellent enantionselectivities (up to >99% ee). The polymeric catalysts were insoluble in commonly used organic solvents. They were easily recovered and reused several times without any loss of the catalytic activity.

Sulfate radical (SO4 center dot(-)) and hydroxyl radical (center dot OH) generated from persulfate or peroxymonosulfate in AOPs have been widely used in contaminant degradation. Anthracene (ANT) can be decomposed by SO4 center dot(-) and center dot OH. The processes of ANT decomposition were investigated using theoretical calculations in this paper. The initiation reactions of ANT, anthrone, anthraquinone (ATQ) and 1-hydroxylanthraquinone (1-hATQ) by two radicals are studied. The highest free energy barriers of initiation reactions are 22.30 kcal mol(-1) in ATQ + SO4 center dot(-) reaction and 6.84 kcal mol(-1) in ATQ + center dot OH reaction. Comparing the rate constants of initiation reaction through the two radicals at 273-373 K, it can be concluded that SO4 center dot(-) and center dot OH both play important roles on the initiation of ANT and anthrone at lower pH. For ATQ and 1-hATQ, center dot OH is more important than SO4 center dot(-) in the initiation process, which indicates that the indirect influence of SO4 center dot(-) are more significant in the degradation processes of ATQ and 1-hATQ. This study provides theoretical confirmations for the mechanisms of reactions of ANT with SO4 center dot(-) and center dot OH, and evaluates the importance of SO4 center dot(-) and center dot OH according to the reaction rates. The work can give more insight into the degradation of PAHs by radicals.

During the past few decades, fabrication of functional rotaxane-branched dendrimers has become one of the most attractive yet challenging topics within supramolecular chemistry and materials science. Herein, we present the successful fabrication of a family of new rotaxane-branched dendrimers containing up to 21 platinum atoms and 42 photosensitizer moieties through an efficient and controllable divergent approach. Notably, the photosensitization efficiencies of these rotaxane-branched dendrimers gradually increased with the increase of dendrimer generation. For example, third-generation rotaxane-branched dendrimer PG3 revealed 13.3-fold higher O-1(2) generation efficiency than its corresponding monomer AN. The enhanced O-1(2) generation efficiency was attributed to the enhancement of intersystem crossing (ISC) through the simple and efficient incorporation of multiple heavy atoms and photosensitizer moieties on the axles and wheels of the rotaxane units, respectively, which has been validated by UV-visible and fluorescence techniques, time-dependent density functional theory calculations, photolysis model reactions, and apparent activation energy calculations. Therefore, we develop a new promising platform of rotaxane-branched dendrimers for the preparation of effective photosensitizers.

The polysaccharide of yam was extracted by hot water method and purified by column chromatography. The physicochemical properties of Chinese yam polysaccharide were analyzed by UV, IR, GPC, 1D-NMR and 2D-NMR spectra. The results showed that Chinese yam polysaccharide had alpha-D-Gluc-(1 -> 4) glycoside bond, and the C2 hydroxyl group was replaced by ethoxyl group. The average molecular weight was determined to be 7.28 x 104. It showed that The scavenging effect of yam polysaccharide on hydroxyl radicals was similar to VC. The sulfated polysaccharide (S-P), phosphorylated polysaccharide (P-P), carboxymethylated polysaccharide (C-P) and acetylated polysaccharide (A-P) were identified by IR and NMR. The results showed that P and its derivatives showed good antioxidant activity. Especially, their scavenging ability to hydroxyl radicals reached the level of VC. This laid a theoretical foundation for the development of yam polysaccharide-related foods.

We report the synthesis of inexpensive and environmentally benign cobalt(0) nanoparticles on L-3,4-dihydroxyphenylalanine (L-dopa) functionalized and magnetite (Fe3O4) grafted graphene oxide (Co@GO/Fe3O4/L-dopa) which was fully characterized with different techniques such as Scanning Electron Microscopy (SEM), High Resolution Transmission Electron Microscopy (HR-TEM), X-Ray Photoelectron Spectroscopy (XPS), X-ray Powder Diffraction (XRD), Thermogravimetric analysis (TGA), Fourier Transform Infrared Spectroscopy (FTIR), Vibrating Sample Magnetometer (VSM), Carbon Hydrogen Nitrogen (CHN) analysis, Energy Dispersive X-ray (EDX) and Inductively Coupled Plasma Atomic Emission Spectroscopy (ICP-AES). Graphene oxide was used as the core material due to its mechanical, electrical and thermal properties. In order to avoid the cumbersome process of separation, magnetite nanoparticles were coated over the graphene oxide. After the successful preparation of graphene oxide based magnetic nanoparticles, L-dopa was grafted over Fe3O4 nanoparticles so as to provide firm anchoring agent for cobalt nanoparticles. Finally, cobalt(0) nanoparticles were immobilized on the developed magnetic support. The catalytic efficiency of the synthesized catalyst was tested for Suzuki cross-coupling and oxidation reactions, usually carried out by precious and expensive second and third row transition metals; products were obtained in good to excellent yields. The synthesized catalyst represents an attractive alternative to conventional catalysts for Suzuki cross-coupling and oxidation reactions, and is recyclable up to five runs.
